| NTCP

Sodium/bile acid cotransporter also known as the Na+-taurocholate cotransporting polypeptide (NTCP) or liver bile acid transporter (LBAT) is a protein that is encoded by the SLC10A1 (solute carrier family 10 member 1) gene.Sodium/bile acid cotransporters are integral membrane glycoproteins. Human NTCP contains 349 amino acids and has a mass of 56 kDa.Bile acid:sodium symporters participate in the enterohepatic circulation of bile acids. Two homologous transporters are involved in the reabsorption of bile acids. One of these absorbs bile acids from the intestinal lumen, the bile duct, and the kidney with an apical localization (ileal sodium/bile acid cotransporter).

| Typical data

As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, wash technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data are provided below for reference only.

Concentration (ng/mL) 50 25 12.5 6.25 3.12 1.56 0.78 0
OD 2.53 1.84 1.13 0.67 0.4 0.3 0.22 0.08
Corrected OD 2.45 1.76 1.05 0.59 0.32 0.22 0.14 -

| Repeatability

Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%

| Recovery

The recovery of Human NTCP spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.

Sample Type Range(%) Average Recovery(%)
Tissue Homogenates(n=8) 89-105 101
Cell Lysates(n=8) 96-108 104

| Linearity

Samples were spiked with high concentrations of Human NTCP and diluted with Universal Diluent to produce samples with values within the range of the assay.

    Tissue Homogenates(n=4) Cell Lysates(n=4)
1 : 2 Range (%) 86-96 90-110
Average (%) 93 96
1 : 4 Range (%) 87-108 105-115
Average (%) 98 110
